Ampicillin and Cloxacillin is a broad spectrum penicillin & formulation of Ampicillin and Cloxacillin by uniting the antibacterial spectra of these complimentary penicillins Ampiclox provides bactericidal activity against an extensive range of gram positive and gram negative bacteria. Most strains of meningococcus. pneumococcus and gonococcus are sensitive to ampicillin. As it is destroyed by B-lactamase, the drug is ineffective in most staphylococcal Infections. The drug has no other clinical pharmacological activity apart from its antibacterial effect. Streptococcus viridans and Listeria monocytogenes remain sensitive to low concentrations of ampicillin.
Cloxacillin when administered has peak plasma concentration of about 15 mg per ml. 30minutes after a dose of 500mg. About 94% of it is bound to plasma proteins. Cloxacillin crosses the placenta and is excreted in breast milk.
The recommended adult dose is one capsule every six hours, one hour before meals or at least two hours after meals.
Known hypersensitivity to penicillin, patients with either glandular fever or lymphatic leukemia show an increased tendency to develop maculopapular rash. Ampicillin should not be used where the etiological agent is known to be ampicillin resistant.
Patients sensitive to penicillin should be given an antibiotic of another class . Care is necessary while administering high dose of penicillin, especially if renal function is poor because of the risk of neurotoxicity.
As with all penicillin, there is a small possibility of anaptiylaxis , which is more likely with parenteral than oral administration Ampicillin is also known to give hypersensitivity reactions from urticaria and fever to generalized anaphylaxis. Convulsions occur in patients prone to seizures after administration of normal doses of ampicillin and cloxacillin . Hypersensitivity myocarditis causing cardiac failure has been reported but is very rare. Acute pancreatitis is a rare complication of ampicillin treatment. Anaptiylaxis to need anaphylaxis.
Hepatitis and cholestatic jaundice have been reported occasionally with cloxacillin and may be delayed in onset of phlebitis.
Patients sensitive to penicillin should be given an antibiotic of another class . Care is necessary while administering high dose of penicillin, especially if renal function is poor because of the risk of neurotoxicity.
